Definition

Let X1,…,XnX_1, \dots, X_n be i.i.d. real random variables with a continuous distribution, and let mm be its median. Let m0∈ℝm_0 \in \R. The sign statistic is S=#{i:Xi>m0}.S = \#\set{i : X_i > m_0}. The sign test of H0 ⁣:m=m0H_0 \colon m = m_0 uses the fact that under H0H_0 the statistic SS has the binomial distribution with parameters nn and 1/21/2. Against H1 ⁣:m>m0H_1 \colon m > m_0 it rejects H0H_0 for large SS, against m<m0m < m_0 for small SS, and against m≠m0m \ne m_0 for SS far from n/2n/2.

Remarks

For matched pairs (Yi,Zi)(Y_i, Z_i) one applies the test to the differences Xi=Yi−ZiX_i = Y_i - Z_i with m0=0m_0 = 0. Continuity of the law gives P(Xi=m0)=0P(X_i = m_0) = 0; with real data, observations equal to m0m_0 are usually dropped and nn is reduced.

Example: n=10n = 10 observations, all above m0m_0, so S=10S = 10. The one-sided p-value is P(S≥10)=2−10≈0.001P(S \ge 10) = 2^{-10} \approx 0.001 under H0H_0, and H0H_0 is rejected at level 0.050.05.

Non-example: let P(Xi<m0)=P(Xi>m0)=1/4P(X_i < m_0) = P(X_i > m_0) = 1/4 and P(Xi=m0)=1/2P(X_i = m_0) = 1/2. Then m0m_0 is a median, but the law is not continuous, and SS has law Bin(n,1/4)\mathrm{Bin}(n, 1/4) instead of Bin(n,1/2)\mathrm{Bin}(n, 1/2).

The Wilcoxon signed-rank test also uses the sizes of the deviations Xi−m0X_i - m_0, at the cost of assuming symmetry.
